XPure-Systems plans to develop a flexible simulated moving bed (SMB) process for purifying target molecules from biological feed streams, such as fermentation broth or anaerobic digestion. Ion-exchange (IX) has been identified as a suitable separation technique, with the IX step performed in SMB mode for continuous processing. One specific operational mode is Expanded Bed Adsorption (EBA), where IX chromatography columns are used in upflow.
The described SMB system is designed for experimental use, with experiments typically lasting 8-24 hours and minimal attendance required. The system features a modular valve block with dedicated inlet and outlet valves for each column, supported by peripherals including pumps, sensors for temperature, pressure, UV, pH, and conductivity, and options for fractionation. The automation level is sufficient to:
- Execute process conditions via online recipes with minimal attendance
- Run multiple configuration recipes
- Log and retrieve relevant process data
- Use process data in real-time for process control
The User Requirements Specification (URS) aims to list all user requirements for the level control subsystem in the XPure-S expanded bed SMB mode. The system will be used for multi-column chromatography processes based on expanded bed adsorption and for handling turbid streams containing biological material as suspended solids. The URS scope includes a control system capable of regulating liquid flow rate, utilizing an ultrasound level control system among other methods.
